Upper Wimpole Street forms the northern continuation of Wimpole Street, running from New Cavendish Street towards Marylebone Road, in the W1G postal district. The designation of an upper portion reflects the street's extension northward as the Marylebone grid was completed during the late eighteenth and early nineteenth centuries, with the layout following the general Howard de Walden Estate plan for the district.

The street shares the medical character of the broader Harley Street district. A blue plaque at number 20 commemorates Ethel Gordon Fenwick, the pioneering nursing reformer who lived and worked there with her husband, Dr Bedford Fenwick. Fenwick campaigned for the state registration of nurses in Britain for over thirty years, a cause that was not achieved until the Nurses Registration Act of 1919. Her presence on this street sits within a broader tradition of healthcare professionals resident in the Wimpole and Harley Street corridor.

Architecturally, Upper Wimpole Street presents a run of late Georgian and early Victorian terraced properties, many of which have been adapted over time for medical consulting use. British Listed Buildings records include terrace sections at numbers 14 to 23. The Royal Society of Medicine, with its entrance at 1 Wimpole Street, is close by. The street has a quieter character than the main spine of Harley Street, while remaining entirely integrated into the medical district that defines this corner of Marylebone.
